RESUMO

OBJECTIVE: To find a way and form of presenting impedance metrics data to clarify prescription of death coming in a comprehensive assessment. MATERIAL AND METHODS: The impedance of biological objects was measured depending on prescription of death coming in standardized conditions (air temperature +4 °C, humidity 45%). The main examined postmortem period was 1 months. Measurements were made on 3 diagnostic zones of biological object and for 5 current frequency of the study. A total of 2100 readings were taken. RESULTS AND CONCLUSION: The studies performed in standardized conditions have showed that using impedance metrics method to diagnose prescription of death coming is possible. The analysis of only absolute impedance values is not sufficient to clarify prescription of death coming due to its undulating changes. The additional use of relative coefficients, characterizing impedance dispersion, is suggested. The study of possible using the impedance indicators of corpse's tissues to determine prescription of death coming should be performed with consideration to pathomorphology of degradation. Further study of impedance dynamics when corpse is under different conditions is necessary for development of a working algorithm to determine prescription of death coming based on tissue impedance.

RESUMO

The aim of the scientific work is to analyze the prospects of using the impedance monitoring method to determine the prescription of death coming. The conducted exploratory analysis allows hypothesizing the dependence of impedance values and dispersion factors for the diagnostic zones studies on the postmortem interval; it also suggests that it is possible to determine this interval for the objects (pig corpses) studies with a combination of impedance values and the corresponding factors. Among large mammals, a swine is the closest to a human from the point of analyzing the postmortem period process, and it can be recommended as an adequate human corpse model. When determining the correlation between the postmortem interval and the impedance parameters, it is the simplicity and reproducibility of the method, the absence of expensive equipment, its portability and the rapid receipt of results that allow this method to be applied at the scene, reinforcing traditional forensic methods for determining the prescription of death coming. Interpretation of impedance monitoring results can be used to analyze the biological nature of the postmortem period processes.

RESUMO

THE AIM OF THE STUDY: Is to justify using the corpses of large mammals as model objects for studying the postmortem period. Similarities in processes occurring postmortem in human and swine corpses (decomposition stages and the structure of dominant necrophilic organisms inhabiting the corpse), as well as similar changes of relative impedance parameters for the cartilaginous tissue and musculoskeletal flap of swine and human corpses have been established. The results obtained allow recommending the swine corpse as an adequate human corpse model both for scientific studies and for solving specific scientific and practical issues arising in the practice of forensic examination when determining the prescription of death coming and the postmortem period conditions.

RESUMO

A model of a chronic lung inflammation in SPF Sprague-Dawley rats was developed by repeated intratracheal administration of LPS in a dose of 0.4 mg/kg. On day 22 of the study, male rats treated with LPS have relative monocytopenia and reduced mean concentration of hemoglobin in the erythrocyte and the mean platelet volume in comparison with the control animals (saline). Intratracheal administration of LPS induced an inflammatory process in the lungs characterized by focal atelectasis, compensatory emphysematous expansion of subpleural pulmonary acini, focal mononuclear and neutrophilic perivascular and peribronchial infiltration, and minor focal mononuclear and neutrophilic infiltration of the alveolar walls. Against the background of LPS administration, germinal centers appeared in the lymphoid follicles of the white pulp of the spleen, and focal mononuclear infiltration of the tracheal mucosa and/or submucosa was observed in some animals.

INTRODUCTION: Pompe disease (PD) is a rare metabolic myopathy with an ample and heterogeneous clinical spectrum, particularly late onset PD (LOPD), which is characterized by appearance at older age and slower disease progression, leading to diagnostic confirmation difficulty and delay. AIM: To describe the genotype and clinical characteristics of Mexican patients with LOPD. MATERIAL AND METHODS: Clinical information from 19 Mexican patients with LOPD confirmed with enzyme activity and GAA gene analysis was reviewed. Genetic information of our population was crossed with international genetic databases. RESULTS: Median age between onset of symptoms and diagnosis was 19 years (range 2-43) and diagnostic confirmation 36 years (range 9-52). Most frequently referred symptoms were proximal axial weakness (n = 17; 89.5%), waddling gait (n = 17; 89.5%) and hyperlordosis (n = 7; 36.8%). Sixteen patients (84.2%) were evaluated with electromyography; a myopathic pattern was reported in 11 (57.8%), but only in 5 patients (26%) paraspinal muscle evaluation was included. The most pathogenic mutations in our group were c.-32-13T>G, c.1799G>A and c.1082C>T. CONCLUSIONS: Similar to other international publications, LOPD in Mexico is clinically heterogeneous; patients may delay years before diagnosis is established. Axial and proximal weakness is the most frequent clinical feature; thus, electromyography with paraspinal muscle evaluation is essential. Except for one, the mutations found in our patients have been previously reported in PD genetic databases.


TITLE: Enfermedad de Pompe de inicio tardío: análisis de una casuística de 19 pacientes mexicanos.Introducción. La enfermedad de Pompe es una miopatía metabólica rara con espectro clínico heterogéneo, especialmente la de inicio tardío, cuya sintomatología es de progresión más lenta y representa un gran reto diagnóstico. Objetivo. Describir el genotipo y las características clínicas de pacientes mexicanos con Pompe de inicio tardío (LOPD). Material y métodos. Se incluyó a 19 pacientes mexicanos con LOPD confirmada mediante actividad enzimática y estudio molecular del gen GAA. Se evaluaron datos clínicos y se revisaron las mutaciones en bases de datos genómicas. Resultados. La mediana de edad de inicio de los síntomas fue de 19 años (rango: 2-43 años), y la edad de diagnóstico, de 36 años (rango: 9-52 años). Los síntomas más frecuentes fueron debilidad axial y proximal (n = 17; 89,5%), marcha basculante (n = 17; 89,5%) e hiperlordosis (n = 7; 36,8%). A 16 pacientes (84,2%) se les realizó electromiografía; 11 (57,8%) describieron patrón miopático y sólo en cinco pacientes (26%) se incluyó la valoración de los músculos paraespinales. Las variantes patogénicas más frecuentes en nuestra casuística fueron c.-32-13T>G, c.1799G>A y c.1082C>T. Conclusiones. Parecido a lo comunicado en publicaciones internacionales, la LOPD en México es clínicamente heterogénea; los pacientes pueden tardar años en llegar al diagnóstico. La debilidad muscular axial y proximal es el dato clínico más frecuente, por lo que la electromiografía debe incluir valoración de los músculos paraespinales. A excepción de una, las mutaciones encontradas en nuestra serie de casos se encuentran previamente descritas en las bases de datos de enfermedad de Pompe.

The use of a high-fat diet, along with streptozotocin administration, can provide more profound insight into the mechanism of development of complications in diabetes, as well as their treatment. High-fat diet given over 3 weeks before intraperitoneal injection of streptozotocin in a dose of 40 mg/kg promoted the appearance of hyperglycemia in Wistar rats. The biochemical analysis of blood serum revealed increased levels of urea, triglycerides, cholesterol, AST, ALT, and concentration of inorganic phosphates and K+ ions in the high-fat diet group in comparison with the control. Both the biochemical analysis of the blood and histological analysis showed more pronounced abnormalities in rats receiving high-fat diet in comparison with animals receiving standard ration. These changes are the early markers for the development of nephropathy, impaired liver function, and microvascular disorders typical of patients with diabetes mellitus.

Objective - to compare the temporal dynamics of the tissue impedance of human and pig corpses under the same conditions to substantiate the further use of pig corpses as model objects. At the first stage, we studied the tissue impedance values obtained on 4 pig cadavers with a known date of death measured with a Keysight U1731 parting. The necessity of using a model object for studying the dynamics of impedance in the postmortem period has been substantiated. An exploratory analysis was carried out that allows to put forward a hypothesis about the existence of the impedance values (absolute indicators) and dispersion coefficients (relative indicators) dependence on the time of death, and also suggests that the combination of impedance values and the corresponding coefficients can be used to judge its duration for the objects under study.

The purpose of this work is to establish the dynamics of the electrical conductivity of various corpse tissues using impedance measurements to assess this method for diagnosing the time of death. It was studied the values of the tissue impedance from the corpses of 8 persons of both sexes and different ages (30-50 years) who died from various causes, with the known date of death. A Keysight U1731 parting was used with two probes, which are stainless steel needles with a diameter of 0.5 mm and a 5 mm immersion length. The analysis of special literature on using indicators of the electrical conductivity dynamics of the corpse tissues, determined by the method of impedance measurement, in the early and late postmortem periods to diagnose the time of death. Trial measurements of a methodological nature were carried out with diagnostic zones: skin, cartilage tissue and tendon. The impedance-metric approach is promising for the purposes of forensic medical diagnostics of the time of death; however, it requires more careful experimental work to establish the dynamics of changes in the total electrical resistance of various tissues of a whole corpse over a long period from the moment of death.

Objective To evaluate prospects for clinical use of circulating biomarkers for characterizing fibrotic changes in the myocardium of patients with hypertrophic cardiomyopathy (HCMP) with left ventricular (LV) outflow tract obstruction.Materials and Methods This was a prospective study with a 12-month follow-up period. The study included 47 patients (29 females and 18 males) with obstructive HCMP who were selected for septal reduction. Echocardiography (EchoCG), cardiac magnetic resonance imaging (MRI) and measurements of serum C-reactive protein, N-terminal pro-brain natriuretic peptide, and relevant circulating markers of fibrosis (TGF-ß1, MMP-2,-9, TIMP-1, galectin-3, sST2, CITP, PICP, and PIIINP) were performed for all patients. All patients were evaluated at baseline and at 7 days, 6 and 12 months following surgical treatment. Morphometrical analysis of intraoperative biopsy samples was performed for evaluation of the degree of fibrotic changes. Patients received beta-blockers (95.7%), angiotensin-converting enzyme inhibitors/angiotensin receptor blockers (34%), loop diuretics (68.1%), aldosterone antagonists (34%), and statins (66%).Results Women with HCMP were older and more frequently had additional risk factors (arterial hypertension). Men had a higher risk of sudden cardiac death. Histological study of intraoperative myocardial biopsy samples showed that the area of fibrotic changes was 13.9±6.9%. According to cardiac MRI mean area of delayed contrast enhancement was 8.7±3.3% of LV myocardial mass. No association was established between traditional cardiovascular risk factors and severity of myocardial fibrotic changes or levels of circulating fibrosis markers. Perhaps that was due to the modifying effect of the drug therapy received by HCMP patients. According to EchoCG maximum pressure gradient in the LV outflow tract before the surgical treatment was 88 (55; 192) mm Hg, and interventricular septal thickness was 22 (16; 32) mm. A considerable decrease (p=0.0002) in the LV outflow tract gradient was observed after myectomy in all patients. At the same time, the left ventricular dimension, which tended to decrease in the early postoperative period, returned to baseline values by the 6th month of follow-up.Conclusion The study confirmed the increase in relevant circulating markers of fibrosis in patients with obstructive HCMP. At the same time, no correlation was observed between levels of circulating biomarkers and severity of fibrosis according to data of histology and cardiac MRI, which was probably due to the modifying effect of drug therapy and limited sampling.

The favorable responses of crop species to low-dose γ irradiation can help to develop cultivars with increased productivity and improved stress tolerance. In the present study, we tried to reveal the candidate metabolites involved in growth stimulation of barley seedlings after applying low-dose γ-radiation (60Co) to seeds. Stimulating doses (5-20 Gy) provided a significant increase in shoot length and biomass, while relatively high dose of 100 Gy led to significant inhibition of growth. Gas chromatography-mass spectrometry metabolomic analysis uncovered several compounds that may take part in radiation hormesis establishment in irradiated plants. This includes molecules involved in nitrogen redistribution (arginine, glutamine, asparagine, and γ-aminobutyric acid) and stress-responsive metabolites, such as ascorbate, myo-inositol and its derivates, and free amino acids (l-serine, ß-alanine, pipecolate, and GABA). These results contribute to the understanding of the molecular mechanisms of hormesis phenomenon.

The association of branched polyethyleneimine (PEI) with a series of octacarboxy-calixresorcinarenes bearing different low-rim substituents leads to the formation of nanosized supramolecular complexes. The PEI-macrocycle complexes have fine-tunable sizes regulated by variations in the self-association capacity of the calixresorcinarenes. In the supramolecular complexes, hydrophobic fragments of the polymer and calixresorcinarenes form cooperative hydrophobic domains which provide synergistic enhancement of guest molecule binding. The formation of the supramolecular complexes was investigated by NMR FT-PGSE, NMR 2D NOESY, DLS and TEM methods. In addition, fluorimetry and UV-vis methods were used with the help of optical probes, namely water-soluble Crystal Violet and water-insoluble Orange OT. The investigation demonstrates the first example of the formation of cooperative hydrophobic domains in supramolecular polyelectrolyte-macrocycle complexes which enhance the binding of both water-soluble and water-insoluble organic compounds. The presented supramolecular systems have potential as sensory and drug delivery systems.

Here we represent the first example of the formation of mixed nanoscale associates, constructed from amphiphilic calixresorcinarenes and hydrophobic carboxylic acids including drugs. The amidoamino-calixresorcinarene self-associates effectively solubilize hydrophobic carboxylic acids - drugs such as naproxen, ibuprofen, ursodeoxycholic acid and aliphatic dodecanoic acid - with the formation of the mixed aggregates with the macrocycle/substrate stoichiometry from 1/1 to 1/7. The ionization of organic acids and the peripheral nitrogen atoms of the macrocycles with the subsequent inclusion of hydrophobic acids into the macrocycle self-associates is the driving force of solubilization. In some cases, this leads to the co-assembly of the macrocycle polydisperse associates into supramolecular monodisperse nanoparticles with the diameter of about 100 nm. The efficiency of drug loading into the nanoparticles is up to 45% and depends on the structure of organic acid. The dissociation of the mixed aggregates and release of organic acid are attained by decreasing pH.

AIM: Evaluate prevalence of genetic variants of hepatitis B, viruses in population of various regions of Uzbekistan with hepatitis of various genesis and different severity levels of liver fibrosis and cirrhosis. MATERIALS AND METHODS: Blood plasma and liverbiopsy from 39 patients with different severity levels of liver fibrosis and cirrhosis served as study material. Genotyping based on direct sequencing of Pre-S1/Pre-S2/S HBV DNAregion was applied. RESULTS: Hepatitis B virus was detected in 32 samples ofthe 39 provided, frequency of occurrence - 82%, respectively. Phylogenetic analysis has shown, that only genotype D was detected among the examined,patients, hepatitis B virus subtype D1 predominated (84.38%) compared with D2 (3.12%) and.D3 (12.5%) subtypes. CONCLUSION: Large-scale sequencing of HBV in, Central Asia will allow to evaluate routes of transmission and time of evolutionary separation or virus isolates. Understanding the epidemiology of the infectious process is important for development of programs for prophylaxis and therapy of the infection.

AIM: Evaluate significance of covalently closed circular DNA of hepatitis B virus as a marker for detection of occult viral hepatitis B in Uzbekistan population with hepatitis of various genesis. MATERIALS AND METHODS: Blood plasma and liver biopsy from 39 patients with different severity levels of liver fibrosis and cirrhosis served as study material. HBV covalently closed circular DNA detection was carried out according to Pollicino T et al. (2004). RESULTS: Covalently closed circu- lar DNA of hepatitis B virus was detected in 82% of samples, including in 54.5% of patients with chronic viral hepatitis C (CVHC) and in 100% of patients with hepatitis of unknown etiology. Quantitative evaluation of content of covalently closed circular DNA of hepatitis B virus in liver tissue in patients with CVHB has shown an average of 2.5 copies of HBV genome as ccc DNA per cell, in patients with CVHB + D an average of 0.7 copies/cell, in patients with co-infection by HCV and HBV - 0.5 copies/cell, in patients with CVHC an average of 0.12 copies/cell, and in patients with cryptogenic hepatitis - 0.2 copies/cell. CONCLUSION: Detection of HBV DNA is a complex problem for effective laboratory diagnostics of hepatitis. Detection of HBV ccc DNA as a marker of occult hepatitis B in patients with CVHC and patients with hepatitis of unclear etiol- ogy is an. important factor for diagnostics, selection of adequate therapy, prognosis of disease outcome and prevention of development of severe liver diseases.

RESUMO

Polymorphism of glucose-6-phosphate dehydrogenase enzyme was studied in the Scots pine populations growing in the sites of Bryansk region which were radioactively contaminated as a result of the Chernobyl accident. It was revealed that the frequency of mutations in isozyme loci increased along with the level of a dose rate (7-130 mGy/year) in the sites under the study. Significant changes in the activity of this enzyme did not depend on the level of radiation exposure.

RESUMO

A new species of bacteria oxidizing ammonium with nitrite under anoxic conditions was isolated from the activated sludge of a semi-industrial bioreactor treating digested sludge of the Kuryanovo wastewater treatment plant (Moscow, Russia). Physiological, morphological, and molecular genetic characterization of the isolate was carried out. The cells were ovoid (-0.5 x 0.8 µm), with the intracellular membrane structures characteristic of anammox bacteria (anammoxosome and paryphoplasm); unlike other anammox bacteria, it possessed extensive intracellular membrane structures located in layers parallel to the cytoplasmic membrane, but never close to the anammoxosome. The cells formed aggregates 5-28 µm in diameter and readily attached to solid surfaces. The cells were morphologically labile, easily plasmolyzed, and lost their content. Doubling time was 28 days, µ(max) = 0.025 day(-1); optimal temperature and pH for growth were 20-45 degrees C and 8.0, respectively. Phylogenetic analysis of the 16S rRNA gene sequences suggested its classification as a new species of the candidate genus Jettenia (order Planctomycetales). The name Candidatus "Jettenia moscovienalis" sp. nov. was proposed for the new bacterium.

RESUMO

The reaction of barley seeds (Nur and Grace varieties) to γ-irradiation in the dose range of 2-50 Gy was studied. The length and weight of a root, the length of a seedling and germination rate were investigated. The dose range in which we observed stimulation of plant development was evaluated. It was shown that the increase of root and seedling sizes after irradiation of seeds at stimulating doses is associated with the rise in the developmental speed, rather than with their earlier germination. Also the effects of a dose rate, a quality of seeds, humidity and a period of storage on the manifestation of radiation exposure were studied.

UNLABELLED: The aim of the investigation is to identify opportunities electrogastroenterography (EGEG) in the survey and individualization treatment of children with inflammatory diseases of the gastrointestinal tract. MATERIALS AND METHODS: We examined five children 10-15 years apparatus Gastroskan-HEV standard overlay electrodes. All children were treated with chronic gastritis in hospital. This diagnosis was confirmed by fibrogastrocopy. The study was conducted in the morning after fasting for 10-12 hours hunger for 40 minutes, and starting in 5-6 minutes after application of the electrodes. Investigated the maximal fluctuations in the range 3.5-4.2 and minimal changes. RESULTS: All children identified acceleration motor-evacuation function of the gastrointestinal tract on the background of hypersecretory reaction. CONCLUSION: EGEG method is noninvasive, has no contraindications and is well tolerated, allowing multiple examine patients regardless of their age and the severity of the condition and correct the assigned therapy.

The lipid composition of the microbial community inhabiting activated sludge in a pilot reactor for the anaerobic oxidation of ammonium (anammox) at the Kur'yanovo Treatment Plant (Moscow) has been studied. The fatty acid composition is mostly based on common fatty acids C14-C18 (95%) with both normal and isomeric structures. The biomass of activated sludge was found to contain lipids with the so-called ladderane substances (ladder alcohols and fatty acids) that are common for anammox bacteria: C20-[3]-ladderane and C20-[5]-ladderane alcohols and C18-and C20-[3]-ladderane and C18- and C20-[5]-ladderane acids. In addition, the native extract contained both simple and compound ethers of the above-mentioned substances with residues of phosphocholine, phosphoethanolamine, and phosphoglycerine. The spectra of the electron impact and tandem mass spectrometry of certain substances have been obtained and published for the first time.

RESUMO

A new genus and species of bacteria capable of ammonium oxidation under anaerobic conditions in the presence of nitrite is described. The enrichment culture was obtained from the Moscow River silt by sequential cultivation in reactors with selective conditions for anaerobic ammonium oxidation. Bacterial cells were coccoid, -0.4 x 0.7 mm, with the intracellular membrane structures typical of bacteria capable of anaerobic ammonium oxidation (anammoxosome and paryphoplasm). The cells formed aggregates 5-25 µm in diameter (10 µm on average). They were readily adhered to solid surfaces. The cells were morphologically labile, they easily lost their content and changed their morphology during fixation for electron microscopy. The organism was capable of ammonium oxidation with nitrite. The semisaturation constants Ks for nitrite and ammonium were 0.38 mg N-NO2/L and 0.41 mg N-NH4/L, respectively. The maximal nitrite concentrations for growth were 90 and 75 mg N-NO2/L for single and continuous application, respectively. The doubling time was 32 days, µ(max) = 0.022 day(-1), the optimal temperature and pH were 20 degrees C and 7.8-8.3, respectively. According to the 16S rRNA gene sequencing, the bacterium was assigned to a new genus and species within the phylum Planctomycetes. The proposed name for the new bacterium is Candidatus Anammoximicrobium moscowii gen. nov., sp. nov. (a microorganisms carrying out anaerobia ammonium oxidation, isolated in the Moscow region).
